Start with a Low Mag. objective (such as 10x or 20x). Setting the DAPI or GFP channel, make coincide the center of the slide with respect to the objective. Adjust focus through the eyepiece. Then move the slide in order to observe the patterns and move to the objective of interest.
Find a repositioning cross pattern and adjust your acquisition parameters on it. Once set, move the pattern of interest.
Do not illuminate with irradiances (peak or average) higher than 50 GW.cm$^{-2}$
Do not zoom in inside a pattern, this could damage the pattern. The area of the scanned zone should not be smaller than the area of the pattern (except for the field of rings - see user guide).
Exposure to water for more than 20 minutes in a row should be avoided. Use oils with the same refractive index as water as an immersion liquid.
After each use, with lens tissue and alcohol only. Do not use acetone.
