Daybook Analysis – Version 2.0.0 – October 2024

The quality of microscopy images is of great importance when these images are to be subsequently processed by analysis algorithms to extract quantitative parameters. Therefore, particular care must be taken during the acquisition and the saving of microscopy images.

Considerations regarding image acquisition precautions and image properties are described in this documentation, to help users of Argolight and non-Argolight products to correctly acquire and save their images, so that they can easily be processed with Daybook Analysis.

I. EXTRINSIC PARAMETERS AFFECTING THE QUALITY OF AN IMAGE

1. SAMPLE CONDITION

2. MICROSCOPE CONDITION

3. MICROSCOPE MISALIGNEMENT

4. STAGE OR SAMPLE TILT

5. CAMERA PIXEL ARTIFACTS

6. SCANNING SYNCHRONISATION

II. INTRINSIC PARAMETERS DETERMINING THE QUALITY OF AN IMAGE

1. WHAT ARE SIGNAL, NOISE, AND BACKGROUND

2. SIGNAL-TO-BACKGROUND RATIO

3. SIGNAL-TO-NOISE RATIO

4. IMAGE INTENSITY

5. IMAGE DYNAMIC RANGE
